Employing fluorescence in situ hybridization (FISH) and targeted next-generation sequencing (NGS), this study aimed to characterize a series of 10 instances of SCD34FT.
The study enrolled seven men and three women, whose ages ranged from 26 to 64 years. Eight instances of tumors were noted in the superficial soft tissues of the thigh, with one each in the foot and back. The size of these tumors ranged from a maximum of 15 cm to a minimum of 7 cm. Cells, plump, spindled, or polygonal, with glassy cytoplasm and pleomorphic nuclei, were arranged in sheets and fascicles to form the tumors. No noticeable mitotic activity was present, or it was extremely low in quantity. Foamy histiocytic infiltrates, myxoid changes, peripheral lymphoid aggregates, large ectatic vessels, arborizing capillary vasculature, and hemosiderin deposition were among the common and uncommon stromal findings. The predominant gene alterations observed were TP53 (53%) mutations, RHOA (13%) mutations, and KRAS amplification (13%); in contrast, KRAS, BRAF, and PIK3CA mutations were not present. Crohn's disease was a significant factor in the occurrence of 80% of SB-PCCs, including RHOA-mutated cases with a histology differing from SRC types, and a notable appendiceal-type low-grade goblet cell adenocarcinoma (GCA)-like characteristic. Mobile genetic element SB-PCCs presented with high microsatellite instability, or mutations in IDH1 and ERBB2 genes, or FGFR2 gene amplification (one in each instance) on infrequent occasions. This suggests the existence of established or promising therapeutic targets within these aggressive cancers.
